Expression of prM/ E Protein of Dengue Virus Serotype 3 in E. Coli as a Sub Unit Recombinant Vaccine Candidate

Abstrack:

Dengue virus (DV) infection is an endemic disease in tropical and subtropical countries especially Indonesia. Secondary infections with different serotype of dengue virus may cause severe clinical manifestations such as dengue hemorrhagic fever (DHF) and dengue shock syndrome (DSS). Vaccine development is the promising approach for DV infection prevention. Challenge associated with development of tetravalent subunit vaccines led many researches to improve recombinant vaccines for overcoming this disease. Pre membrane and envelope genes (prM/ E) have been reported as an immunogenic antigen. Several studies revealed that prM/E could give higher immune responses than either structural or non structural proteins. Development of subunit vaccines base on Indonesian local circulating DV is challenging. In this study, prM/ E genes of DENV serotype 3 will be cloned into pET-32b and transforms into E. Coli DH5? then expressed in E. Coli BL21. Western Blot method will be used to detect prM/ E protein and predict epitomes to B cell and T cell using B Cell Epitome Prediction Tools and Genetyx. read more

Production and Effectiveness Experiment: LIQUID FERTILIZER FROM SLUDGE OF BIOGAS INSTALLATION- ENRICHED BY (Pseudomonas fluorescens) AS BIO-PROTECTANT

Abstrack:

Liquid sludge byproduct of biogas installations is highly potential to be used as a liquid fertilizer. Liquid sludge can also be used as an alternative culture media for growing the Pseudomonas fluorescens bacteria that can act as bioprotectant for plants. This study aimed to assess the effect of slaughterhouse waste (abattoir) cattle blood in the content of N, P, and K of the liquid fertilizer obtained from the liquid sludge enriched by bioprotectant. This study also examines the growth of the colony population and the effectiveness of antagonistic Pseudomonas fluorescens bacteria to control plant pathogen Fusarium oxysporum fungal in liquid sludge culture media which is enriched by abattoir beef blood waste of different concentrations.The experiment was done four treatments, namely the addition of abattoir beef blood waste 0, 2.5%, 5% and 7,5% was applied. Each treatment was repeated three times except for plant trials in testing the effectiveness of antagonistic bacteria on each treatment which was repeated ten times. The variables observed were: total nitrogen, total phosphorus, total potassium, the growth of the colony population, and the effectiveness of antagonist microbia, Pseodomonas fluorescens. To determine the effect of treatment, the data were analyzed using ANOVA. Duncan Test was applied to examine the difference among the treatment. The results showed that the sludge byproduct of biogas installations that have been used as a culture media for Pseudomonas fluorescens bacterial with additional abattoir beef blood waste 5% and 7,5% organic liquid fertilizer is a good source of N, having average N content of over than 3%. Abattoir beef blood waste concentrations is added to liquid sludge positive influence on linier growth rate of Pseodomonas fluorescens bacterial colonies population. Optimum results in the growth rate calculation of Pseudomonas fluorescens bacterial colonies population on the liquid sludge ware 27,12 Cfu / ml.hour that occurred at the time of 4 hours. Pseudomonas fluorescens bacteria were grown in liquid sludge byproduct of biogas installations with or without the addition of abattoir beef blood waste has the ability to suppress the growth of Fusarium oxysporum fungal both in vitro and in vivo in the greenhouse. read more

Farmer?s Adoption of Sorghum Farming in Marginal Land as Alternative Food Source in Bantul Regency Yogyakarta Province

Abstrack:

Based on the condition of limited of land for agricultural activities to fulfill the food needs especially in Bantul Regency Yogyakarta Province, it is needed an innovation that can support the various agricultural activities. The innovation will be more focused on using of marginal land for sorghum cultivation to improve the productivity of land. So far, villagers have been utilizing home garden for various farming production in micro scale for the domestic needs. In addition to home garden, land resources are also available surrounding the village such as land along river bank and sandy land in coastal area can be utilized for agricultural production in Bantul Regency. The aims of this research are (1) To determine the adoption technology in sorghum farming by farmers in marginal areas, Bantul Regency (2) To determine the diffusion technology in sorghum farming by farmers in marginal areas, Bantul Regency and also (3) to identify various problem and to find problem solving faced by farmers. This research operationally was conducted by survey method. Sampling technique was done by purposive sampling by choosing the farmers who have been experiencing in sorghum cultivation sorghum in Bantul Regency. The research result showed that the adoption?s farmer cultivation of sorghum in marginal land up to 75,21%, process diffusion to transfer innovation using agent of change/extension officers, farmer group, and group leader and the last is the problem solving for sorghum cultivation there is availability land can be used marginal land such as land along river bank, sandy land in coastal area, and home garden; water availability can be solve with sorghum cultivation because it?s needs less water and sandy land to cultivation of sorghum; choice red varieties sorghum to avoid attract birds; underdeveloped market can be arranged by a cooperation with food industries; processing of product could create the value added; and giving skill to women farmer to make several product domestic for flour sorghum. read more

Mechanical Water Purification Technology to Provide Clean Water in an Emergency

Abstrack:

Natural disasters always pose problems for humans and the environment. In the unusual natural conditions such as floods, devastating earthquakes, volcanic eruptions, clean water to its own problems for humans. The earthquake and tsunami in Aceh on 2004 and earthquake in Yogyakarta on 2006 are the clear example of environmental damage caused by natural cause an emergency situation. The purpose of this research include generating appropriate technologies, such as water purification equipment with mechanical energy to provide clean water in an emergency, produces environmentally friendly technologies as an effort to improve the development environment (sustainability development). This study is divided into two phase, the first phase is the generation of electric power. The 12 Volt dynamos are driven by a paddle wheel bike R.30, to fill accumulator 12 Volt DC after rectified with Diode Bridge. Accumulator is used as a driving force of water pump after invertu singin verter 12 Volt DC-AC. The second phase is water purification device. The first serves as a coagulation box has a filter Bioform 4cm. The second box serves as achlorination by 4cm Bioform filter and charcoal, while the third has a filter Bioform 4cm and charcoal serves as a water tank. Coagulant dose ratioused 0.50 grams / literalum, 0.1grams/liter of limes tone with a contact time of 1 hour, while the chlorination process using a dose of 0.05grams/liter of chlorine. For 5 minutes. Sample data collected from several rivers flowing in the region of Yogyakarta province. The parameters tested include Total Suspended Solids (TSS), Turbidity (NTU), the content ofiron (Fe), manganese content (Mn), Sulfate (SO4), Degree of acidity (pH) and Total Coliform. Results of samples tested at Health Laboratory Yogyakarta. Mechanical water purification technology using a bicycle wheel radius (R) 30 cm equipped 12 Volt dynamo as a source of electrical energy, Diode Bridge 1 Ampere, the accumulator 12 Volt 5 Ah, 12 Volt Inverter DC-AC, 16 Watt mini water pump as well as a series of coagulation bath, chlorination and water tank is equipped bioform 4 cm and charcoal, can be used effectively for the provision of clean water in an emergency. Coagulant dose ratio of 0.50 gram / liter alum, 0.1 grams / liter of limestone and 0.05 grams / liter of chlorine can lower water levels on the parameters TSS (Total Suspended Solid), Turbidity (Ntu), Iron (Fe), manganese (Mn), pH and Total Coliform in accordance with the water quality standards required. read more

Utilization of Exhausted Gas of Engine Generator to Drive Biogas Compressor

Abstrack:

Availability of fossil energy sources is now limited due to high exploitation until now. Biogas is one of alternative energy, which may be developed to solve the energy problem. Biogas is product of the degradation of organic material through the anaerobic digestion process. Biogas consists of methane, carbon dioxide, water vapor, hydrogen sulfide and other gases. Biogas may be used as fuel in a combustion engine as the driving in electrical generators. Supply pressure of biodigester is usually low, so compressor is needed to increase pressure and stabilize the gas supply to the engine generator. Turbocharger which utilizes exhaust gas of Genset can be used to compress biogas. Specification of generator set rated power of 2000 watts (2200 watt peak) with a gasoline engine driver 5.5 HP single cylinder capacity 163 cc. Some adjustments needed in the generator set are on (1) compression ratio, (2) carburizing / mixer, (3) adjusting the spark plug gap, (4) the ignition timing and (5) other necessary treatments. Results show, that the turbine speed achieved 70,000 rpm during cold test using pressurized air 7 bars. A compressor output pressure 65 cm-water-column is achieved during unloaded hot test by engine speed of 4000 rpm. Results of test with biogas fuel show, which output power of generator, achieved 1700 W by test using turbocharger and 1200 W without turbocharger.. read more

Sambi Tourism Village is a village which has fascinating exotic tourism. This tourist village at the foot of Mount Merapi, the air is cool and beautiful scenic. This village has a rural sense characteristic, so Sambi can develop into a tourist village. Sambi located in Disaster Prone Region I of Mount Merapi (Kawasan Rawan Bencana I Merapi). It has certain implications related to the impact of the eruption of Mount Merapi. Eruption in October 2010 impacted on the continuity of the activities in the village. These impacts include the destruction of rice field areas caused by debris flow at Kuning River and that tipped at Merapi and Volcanic Ash that resulted in the destruction of agriculture in the village. As a tourist village, Sambi has positive implications, such as road construction and improvement of the people’s economy. From JKT48, it could be seen that women have opportunity in the early age to chase their dreams ? all of indicators of Indonesia MDGs third goal has been accomplished here. However JKT48 is more like a new trick of neocolonialism because (1) JKT48 is 48 brand from Japan, and (2) because of its franchise format, JKT48 must (be) act (-ed) just like their sisters in Japan (AKB48, etc.). JKT48 Members are labors, that are commoditized (and once again subjected) to fulfill (Japan?s) neocolonialism. The Complexity of Wayang Wong Performance and Its Dynamic Artistic Development

Abstrack:

This paper aims to examine the practices and performances of wayang wong ? an art form of Javanese shadow puppet performed by humans in forms of drama and dance ? in relation to the reinforcement of practitioners? developments. Traditionally, wayang means shadow, and wong means human. On- stage performance presents the beautiful aspects, but, off-stage practices remain compelling and dynamic to observe. I build on Judith Hamera?s (2007) work on a dancing community that provides a space for artistic cultural development through creative learning process. Fieldwork was conducted over nine months (November 2012 ? July 2013) that included direct and participant observations to Panca Budaya community, Yogyakarta. In-depth interviews were also deployed for data collection. This paper will analyze how the work of wayang wong creates new social arrangements as well as how it improves developments as articulated by the actors. Analyzing so, I argue that this modified dance drama is purposely re-made for the sustainable survival of wayang wong. read more

Comparative Study of Millennium Development Goals (MDGs) Achievement in Various Regional Economic Typologies in Indonesia

Abstrack:

Economic development in Indonesia has created much kind of regional typologies. Klassen has divided this typology into four namely advanced and rapidly growing region, fast growing region, developed but depressed region and relatively backward region. There are some regional characteristic differences among those typologies. These differences have occurred in many sectors including human resources development sector which can be reflected by differences in the achievement of Millennium Development Goals (MDGs). This research is aimed to determine the achievement of Millennium Development Goals (MDGs) in those regional typologies. In this research, analysis of secondary data uses quantitative methods. Level of MDGs achievement is calculated by the analysis of percentage. Level of MDGs achievement in regional typologies which are used in this research is assessed by doing cross-tabulation between result of regional typology analysis and classification of MDGs achievement. The comparison of MDGs achievement among typologies was analyzed using One Way ANOVA. The results show that most of the provinces in Indonesia have an average percentage achievement of the MDGs under 100%. There are six provinces categorized as region which have a high level of MDGs achievement, 18 provinces have moderate levels of achievement and nine provinces with low levels of MDGs achievement. In the Klassen typology of Indonesia, level of achievement of the MDGs pattern sequence is not corresponding to the sequence pattern of regional economic development level. The order typology with the MDGs levels ranging from the highest to the lowest is developed but depressed region, relatively backward region, advanced and rapidly growing region, and fast growing region. Based on One Way ANOVA test result, there is no significant difference between MDGs achievement in the various typologies. Therefore the difference of regional economic development level in Indonesia did not create a significant difference in the achievement of the MDGs. read more
